Maybe Getting Confused...

Despite enjoying what we've been doing so far in this class, I think I'm beginning to get confused on what our ultimate goal is. It's hard to remember when we did what, too. Maybe we can have a small recap of what we've done every once in a while...I think that would be helpful. Besides that, I think the class and our experiment is going great. I was a little frustrated our DNA extraction didn't work, though. Hopefully that won't affect any other part of our experiment.

  1. Our ultimate goal is to chaarcterize the 78 strains and describe new species of the genera Geodermatophilus, Blastococcus, and Modestobacter. We have been using classical and molecular techniques to do this.Yopur lab note book or the blog tell you exactly "when you did what". The calender in Airset actually tells you when you will read the results from the plates already set up.

I understand what our ultimate goal is but I'm being concerned that due to time constraints we won't be able to run all of the test on the strains that we found in the papers about previously described species. It is absolutely necessary to run all of those test before we can conclusively state that we found a new species? It seems to me that once we have could extract the 16S rRNA and compare it to previously described strains. Is this a ridiculous oversimplification?

  3. I understand now that to describe a new species we have to run all of the test. Many isolates are known simply by 16s rRNA from unknown samples, but the isolates are still undescribed because they cannot be grown or have yet to be grown under laboratory conditions and described with morphological, biochemical, and chemotaxonomic (spelling) test. Got it!
